In this study, the extraction conditions of berberine( alkaloid) from stems of Berbrris vulgaris by subcritical water extraction method were optimized using Response Surface Methodology (RSM). The effects of extraction temperature, time and ratio of sample to water on the concentration of berberine were evaluated. The results showed that all three factors (time, temperature and ratio of sample/solvent) statistically have significant effects on the yield of extracted berberine concentration by subcritical extraction method. The yield of berberine was increased with increasing the temperature and time of extraction condition while the yield was reduced when the solvent – sample ratio was increased .The temperature and ratio indicates that temperature has significant increasing effect while time and sample/solvent ratio have a decreasing effect.The Results Shows that increasing of ratio (sample to solvent), over time, will reduce the concentration of extracted berberine. The curves also show that time have a decreasing effect on Berberine yield
